I would expect the primary route will be through the App Store with DVDs available at retail stores for people with slow / poor internet connections

However it comes, I'd like it installable to a USB key please. Even better if I can upgrade the MBA's original key with it.

I'd buy through the App Store, but I hope for work we can buy physical media (whether that be USB or DVD).

No way they'd sell it on a USB key. Think of the costs compared to a DVD.

I would think they would at least provide the option for the benefit of MacBook Air users. They already distribute it on a USB key for them.

I bet they're conflicted inside Apple over it. While they have the MAS, do they want to give up all that free publicity of people lining up for a new OS release?

Actually, if they use non-rewritable flash (unlike normal USB drives) like the MBA restore drives have, it would be almost the same cost. Shouldn't be an issue.
