Just started my first MKV right now, and I noticed Make MKV recommended that a DASPI kernel extension be installed to improve performance. Does anyone know what this means?

OSX use something call ""OS access mode" which restrict direct access to the optical drives to 3er party software, this could cause some issues when ripping media.

DASPI kernel extension would allow MakeMKV to access optical drive directly, eliminating all problems related to "OS access mode". Installing DASPI driver won't change any system behavior - MakeMKV will not unmount or lock the disc for exclusive access, and disc will be still accessible in finder or any other program.
