How can I make a key combination to pause iTunes from anywhere in Windows? And then use the combination again to unpause it?

Alt+ Tab (till you get to Itunes) then hit the space bar. I know that is not what you are looking for but it is an option.
 
don't most PCs have those ugly multimedia buttons all over the keyboard? Can't you reprogram one of those? Seriously I'm not sure...

I think you get those to load itunes but I don't think you can make it stop/start it.
 
If you have a multimedia keyboard you can use the Mute button (although that will mute the Master volume by default; ie: All sounds in Windows).

Maybe try this software? Called iTunesKeys. Haven't tried it, but might be worth a shot.
 
I'm on a *shudders* windows pc at the moment, and have just opened iTunes. The quickest way that I have found is that you can right hand click on the iTunes icon in the 'tray' at the bottom, and then click 'pause'.
