First post, but I've been lurking for a couple months now. I just got a SR MBP and I'm trying to get rid of my old windows PC. I want to know the best way to back up my MBP's HD on an external HD and also use the External HD for storage (music, video, etc.).

So here's the details:

I have the 160 GB HD in the MBP.
I have a Firewire 800 WD 500GB external Drive.
About 250 GB of Music/movies.

What i'd like to do is create a bootable back up of my MBP's drive on the Ext HDD in one partition and use the other partition for music/movie file storage. What is the best way to do that?

Well, partition your drive with Disk Utility first. Probably 160GB and 340GB. Then use an app like Carbon Copy Cloner (free) or SuperDuper! to clone the main drive onto the smaller partition. Use the larger partition for your multimedia files. The previously mentioned programs can do that backup, too.
