Time Machine Initial Backup

Quick question on Time Machine's initial backup... If I set up time machine for the first time using a firewire 800 connection to my external harddrive and do the first initial backup, will I then be able to disconnect the external harddrive and connect it via USB to my Airport Extreme using AirDisk to continue my normal time machine backups?
