Is is smart practice to wait to the iphone is totally out of battery to give a full charge or charge once it hits say, 20% or even 10% after warning? Also, should you power off the phone when charging or does it not matter?

Well since I've got exactly 2 battery cycles on my new iPhone I can't say what's best, but I've been draining the battery all the way till it shuts off on me then charging it up. Usually I'm also syncing it with my computer so it turns on too for the charge. To track your battery usage you can go to Settings>General>Usage. So far with this charge I have 2h 42m, standby for 1day 1 hour, and 32 minutes call time... Hmm maybe I should start tracking that... :)

It's good to drain down and then fully charge when you first get it to train the battery. Then you can charge it whenever you want after that. It's also good to drain down and fully charge once a month to keep the battery healthy.
If I let my iPhone drain down until it turns off, after a while it turns back on when it has a little bit of power. I haven't tried powering it while it was off since I like being able to get calls while it's charging.
It's OK for it to be on while charging, but it will not charge as fast, since it's on and using power. I haven't checked the difference in charging time for on vs. off.

Well I've heard that charging any battery from 0% too 100% causes more strain on the battery. So if you charged from lets say 50% plus it will cause less strain on the battery cells as there is less to charge.
Obviously though sometimes you can't help the fact that you cant always charge your iphone from +50%
So basically just charge your iphone every night or whenever as much as you can. There is also no need to turn the phone off its perfectly safe to leave it turned on while charging :)

This is that I have done with my original iPhone for the past year, and it is still going strong, with no noticeable loss of Battery life.
Every night I plug it into the dock and charge it up. I leave it on as well, and just take it out in the morning.
So far, it has worked great for me, so I am continuing this process with the 3G as well...
