I've had Panther on for 4 days now and it is quite noticeable that GB performs heaps better.
It uses significantly less memory;
It has very few error messages;
It doesn't beep error will exporting to iTunes;
It rarely gives System Overload messages;
Besides GB, I'm still yet to see enough new stuff in Panther to justify the cost (lucky I got it cheap on eBay) - and Expose hasn't really grabbed me. And I loathe the Windows XP styling of Aqua windows that have focus.
