1TB HD for backup on Macs and Pcs

I have the WD MyBookStudio 1TB external HD and im facing the issue of using it for backup for both my Pc and Mac. What is the optimum way of using it for such a purpose?
 
Format it FAT32. If you need to use files larger than 4GB, format it NFTS and download MacFUSE and NTFS-3G to install on your Mac.
 
So suppose i go with Fat32, what do i do? Do i make 1 big Fat32 partition or make two and why?
