Is it possible to use the speakers that come in the 27in CD as well as the speakers in the 27in iMac at the same time?
 
You can use them at the same time if the application you use supports multiple outputs at the same time. An application that can specify which output to use will also allow you to use a second application to output different audio on the other display.

OS X does not provide anything built in that allows you to route system wide audio to two output devices at the same time. So it is really up to someone to write their application to support specifying outputs outside the current selected "system wide" audio setting.

Also, if you are thinking of using both the iMac and ACD to output the same audio at the same time, I have to tell you that it would sound hideous because the speakers are not the same specs and also it would not be proper synced stereo (would result in an reverb-like effect).

It's just a terrible idea in general. If you want to output separate audio for different purposes though (say, playing music on one and then outputting Skype audio on another) it should be possible if the application supports it.
