Is it possible to backup up multiple Macs via Time Machine to a single FireWire drive without having to unplug and replug the drive into each machine?

No, it is not possible. The two FW800 or FW400 ports are not intended for more than one computer (no this has nothing to do with the so called target disk mode). You can create two or more partitions with different names on your Firewire disk, if you want to save TM backups from different Macs.
