Ethernet cable

Is there any problem if I plug/unplug my Ethernet cable from my iMac 27" (late 2012) (connected to the router which is connected to the cable modem) while system is running or I have to turn off the iMac?

No harm will be done by unplugging the ethernet cable while the machine is turned on.

Besides losing your internet connection.
