Does anyone know if it possible to use MovieTube on an iMac desktop to stream to my TV via Apple TV with AirPlay?

How do I do it then? As I'll start to play a movie and there is not AirPlay icon?

From a Mac you need to mirror the Mac display to the AppleTV. Those controls are in the menu bar and/or in preferences.

You are describing AirPlay (not to be confused with AirPlay mirroring) you'd see on an iOS device (iPhone or iPad) where if it's built into the app you can send the video link to the AppleTV via the AirPay icon.
