In Apple's instructions for calibrating the battery they say that you can either turn the computer off or let it sleep for 5 hours or more. If I turn it off, can I re-charge immediatley or do I also need to wait 5 hours?

Either sleep or off is ok. What they want you to do is not use the computer for 5 hours, but it can be in sleep mode or off while it charges and then calibrates.

Side Note, don't forget to note the make and model and the OS version you have when you ask a question.
