I'd rather have a backup disk separate from a working disk, so 2x500GB disks would be better IMO. Plus it most likely a cheaper solution. If you get a 2 drive enclosure, you could later upgrade to 2x1.5TB drives for futureproofing...

On my 500GB drive, I have one part reserved for Time Machine, the other for media stuff. On the 250 GB, one part is reserved for SuperDuper and the other for miscellaneous.
