I noticed when I updated this afternoon that the file size of 4.0.1 was about twice the size of any previous update. Did anyone else notice this? Why was it so much bigger?

Yup. Any update of the iOS is the who OS, not just a patch.
iOS 4.0 sizes (don't know about 4.0.1):
3G: 292MB
3GS: 377MB
iPhone 4: 579MB

They sent out a whole version of the OS instead of just as patch.

Gotcha. But why was it a whole version? It seems like it would have just been an update for the signal strength. I haven't noticed anything different besides the bar size.

Because it's better to release an OS with the update integrated rather than have it stick onto the OS as a patch. It allows them to keep the OS size around the same.
