Prior to installing Lion I want to ensure that my Boot Drive, which I wanted to store only Applications but in the past have had to store large video files, is as fresh as fresh can get.

I can wipe the Boot Drive clean and then re-install all the Apps and Drivers, and Plug-ins, but this would be a lot of hassle and will be very time consuming.

So is it possible to Copy the boot drive to another internal HD, then install Lion on the Boot Drive whilst wiping the whole Boot Drive clean, and finally copy/transfer back my Apps, which were copied to the other internal HD?

I hope this makes sense :)
