I just purchased a brand new 16gb Wifi (no 3G) from Apple and it has 3.2.2 on it. I might consider jailbreaking it down the road but I want to know if there's anything significant that will happen if I update to 4.2.1 that I might have missed when doing my research.

If you think you may be interested in jailbreaking in the future, currently, 4.2.1 requires a tethered jailbreak. 3.2.2 can be jailbroken untethered.
 
Hrm... ok that's good to know. I like having that switch for what it does. How about speed of the OS, better... worse? Seems lightning fast right now. Folders in the update seems nice but I rarely use it on my iPhone so it's not a big reason for me to update.

Some apps won't work with 3.2.2.
 
try backing up your SHSH blobs in case you ever want to downgrade (if you decide to upgrade)
 
Well I decided I might as well get the device to its latest and greatest format. Jailbreak will come in due time and hopefully at that point it won't be tethered any longer. As to the orientation lock, I completely forgot you can still lock it in the other menu at the bottom.

I got rid of my iP4 to get this and it's gonna be tough not having the portability for the quick trips to wherever but I couldn't stand paying the $30 data any longer when I NEVER used it. So far, very impressed with this device...

Glad you like it. It's a great device. I've never bought anything Apple before but I love the iPad. I must be on it 10-12 hours a day.

Thanks! I had a hard time justifying it with my iPhone4 but now that I've gotten rid of it, this was a must. I've been sucked in since the 2g and now it's time to expand. Might have a MBP coming down the road too...
 
There is now a mod in Cydia tha allows you to revert the mute switch to an orientation lock. I believe it is called Nomute, but you can find it easily.
