Is it possible to purchase AppleCare+ from an Apple store for an iPhone 6 Plus that was purchased from an AT&T store? Haven't had an major issue with my iPhone 6 Plus, but I've been thinking that it's probably better to have AppleCare+ just in case something does happen.

Yes

You will need to purchase it within 60 days of buying the iPhone. Apple will need to verify that it is in working order.
