Under System Preferences, I see that iTunesHelper is set to automatically open when I log on. Is this vital for iTunes to work properly or can I remove it? Would there be a startup time difference?

You can remove it and iTunes will still work properly. The only difference is that it won't automatically open when you plug an iPod/iPhone/iPad by USB.

I doubt it would make a significant boot time difference, but if you don't need your iTunes to automatically open you can remove it, it can only help even if we're talking about a tiny fraction of second.
