Apps that stop working

For some reason Handbrake no longer works on my 24" white iMac. I've tried reinstalling to no avail.
It's happened with other apps once or twice before, and I have no idea what so ever what's wrong, how to find out what's wrong or how to fix it.

I'm running Tiger BTW.

Help!
