I'm hoping to use my wife's upgrade eligibility for purchasing the iPhone 6 on the NEXT plan. Her current contract ends soon, 11/14/2014, so if I pay off the NEXT installment plan to coincide with when her current contract ends, will I be able to successfully get the new iPhone 6 unlocked?

Doubtful. They check the IMEI of the device, too. Also, NEXT installment is not bound by contract. If you pay off NEXT for the iPhone 6, then it's eligible for unlocking whether that's 1 day after receiving the phone or 2 months after.
 
I'm the primary account holder for my wife's phone and can use her upgrade. Her contract ends soon and her iPhone 5S is already unlocked.

Actually, I'm somewhat confused about what you want to do. Do you plan on paying off the NEXT installment plan for the iPhone 6? In that case, you can request an unlock after you pay off the balance. No need to wait for your wife's contract to expire.
