Downloaded a game on my retina iPad Mini via the app store but I wanted the bigger iPad Air so I sold it. I bought the iPad Air and wanted to download the game again from the app store but it was removed by the developers.

Can I backup free and purchased apps to my computer (OS X) so I can put it on my iPad in the future if the developer or Apple removes it/them from the app store?

Yup. In iTunes, plug in your iPad and in the summary there's an option to either back up to iCloud or 'back up to this computer'. Select that and you'll be fine :)

PS bummer about losing your game. I had a friend that happened to and he was gutted.
 
Try going to the "Purchased" tab on the bottom of the App Store and see if it shows up there. I have tons of old apps there available for download even though they're no longer offered for new downloads in the App Store.
