I just recently purchased a 256 MB Sandisk Cruzer USB2 Flash drive. I was wondering what the best format is for such a device. I will be doing a lot of work on both macs and windows machines with the device.

Its current format is MS-DOS File System. Is this the best option for what I want to use it for?

Leave it as it is. Window cannot read native Mac OS formats (HFS, HFS+) and OSX cannot write to NTFS. Fat-32 is the best option in a mixed OS environment as almost everything can read/write it.
