I would like to maintain an off-site backup, and plan to rotate a hard drive on a monthly basis. I'm running 10.5.5 with Time Machine. I understand that I may need to initialize a new backup drive every time I switch drives? Any thoughts? Is Time Machine really the best way to go here?

If you're going to do this, perhaps you'd be better off simply duplicating your Time Machine volume on to the second drive instead of trying to rotate the drives. Time Machine isn't really designed for this.

Consider something like Retrospect if you want a more advanced backup solution.
 
This is a good idea. I have a 2 bay external enclosure, so perhaps I can just pull the drive from the off site, duplicate it at home, and then put it back at the off site. I could do that monthly or bimonthly.
