I want to get an iMac 27" i5 or i7 with 16gb of RAM.

Should I buy it preconfigured with 16gb or is it better to buy the RAM from a third party and add it myself?

Can you upgrade any parts of the iMac?

Yes, you can upgrade the RAM yourself, using RAM from 3rd party vendors, which will be much cheaper than buying RAM from Apple. The User Manual even shows you how.
 
upgrade the RAM yourself...it's very easy and you'll save a lot of money...and you can use those savings towards a SSD ;)
