Can i partition my external my HD to do the following..?

Okay, so i have my MacBook pro, and i have a 320GB WD hard drive. Can i partition the harddrive so that i can have 250GB set for back, and the rest of the 70GB used for just file storage? Meaning that lets say i connect this hard drive to a windows computer to transfer some files from it, it obviously wouldn't see the time machine partition, but will it see the 70GB partition?
