When there was an MP3 on my desktop in a folder, I remember that I would be able to do play it without having to open the larger preview window.

Has this been removed in the recent update (a shame, if so) or is there possibly something I may have done/be doing wrong that is causing it to no longer function as remembered?
 
You can press the SPACEBAR to listen to it in QuickLook or if you have "Icon preview" enabled in the View Options (CMD+J) there should be a little play button inside the clip.

The ability to preview the mp3 in Finder's icon view as you described still works after the 10.6.8 update. Have you tried restarting?
