I have an older USB 2.0 500 gb LaCie drive/enclosure which I'd like to turn into a 3.0 enclosure and drop in a larger 3TB drive.

Is this even possible?
I'm not sure what the cable/bridge is called to change the existing USB 2–>Serial ATA to USB 3.0–>SATA interface.

I realize it may just be easier to buy some cheap 3.0 enclosure, but kind of enjoy tinkering and the LaCie enclosure.
 
maflynn is correct. You'd need an entirely different PCB with an appropriate controller(http://via-labs.com/en/products/vl700/index.jsp) inside that LaCie. And even if you do manage that on the cheap, there's no guaranty of physical compatibility (fit, port alignment, etc.).

Less headache and cheaper to just buy a new enclosure.

Disassemble the enclosure - remove all the electronics - and just Dremel a slot in the back to hold this.
