Can I clone a Mac Mini with 10.6 and completely overwrite an iMac with 10.5 or will OS X 10.6 freak out because it is on a different platform ? If I had to clone the Mac Mini to multiple iMac's what is the fastest way to do it... Firewire with a target drive using Super Duper or USB dongle (or is this even an option ?)

Firewire would be the fastest, and if your Mac OS X 10.6 Snow Leopard install is updated to its latest version (2011), then you can clone that OS to any Intel Mac from 2006 to 2011 (except 2011 MBAs and Mac minis), since it includes all the Macs' drivers.
