I compared the performance of an Ultra 3.5" External Hard Drive Enclosure with different File Systems (FAT32, NTFS, MacHFS+), Interfaces (Firewire, USB2.0), and OSes (Windows XP and Mac OSX); all using a Macbook Core 2 Duo as the test system.

To sum things up, you should use an external enclosure with Firewire and the HFS+ file system unless you require full compatibility with non-OSX operating systems, then you should use Firewire and FAT32.
