Deactivate phone and then back up in iTunes?

Would like to move from AT&T to Verizon for a 4S today, but my wife has not backed up in iTunes in a while (and is using a 3.x version of iOS). Can her phone be deactivated, backed up in iTunes, and then bring a new phone into the fold without risk of losing anything?

She also uses Notes extensively, is there a way to back those up without a lot of manual work?
