Delete my "downloads" for extra HDD space?

Can I delete the content inside my downloads folder? It's full of music and some movies. If I delete the items from this folder will it delete them from iTunes and wherever else they may be?

I'm trying to get some extra space. I've used Onyx, if you know of any other safe ways please let me know.

Use this to find duplicate files. It's safe to delete the Downloads folder if you made copies of the data inside or if you don't want the data anymore.

iTunes can be set to copy media into its own folder, if you want.
