This is probably easy... today when driving home, I played a song and there was a momentary buzz. It was repeatable. When playing the same song on my Mac Pro, there's no problem.

How can I force a re-sync of the functioning file to the iPhone? Or do I need to do a full re-sync (21GB of songs)...

iPhone 3GS (latest ROM as of this timestamp)

I assume that you phone syncs automatically with iTunes? If so, plug into iTunes, select 'manually manage music' then delete the song from your phone. Drag it back over from your library then uncheck 'manually manage music'.
 
Wouldn't it be simpler, in iTunes, to:

1. Select "Sync only checked songs and videos"
2. Uncheck the problem song
3. Sync
4. Check the problem song
5. Sync
