Does greenpois0n require me to do anything in iTunes?

I sync my iPhone 4 to my MBP, but I want to jailbreak my phone here at work, where I have Windows.

Do I need to do anything in iTunes/will I mess anything up by jailbreaking on a different machine than I sync my phone to?

Getting the device into DFU mode instead of recovery mode seems to be the culprit for a lot of people. Are you sure it's in DFU mode?

How can I tell? I'm following the timer exactly as greenpois0n prompts me to...
 
Had similar problems with Greenpois0n (went into DFU mode no problem, but kept getting failures). Limera1n worked perfectly.
