My 3GS defender had a nasty problem of the screen cover sticking to the screen and creating a watermark. I was able to fix it by putting an anti-glare screen protector between the screen and the screen cover of defender. Does this problem still exist for the new iphone 4S defender cases?

My wife has the defender for the iphone 4S and has had no issues with the splotches (she encountered the same problem with her 3GS and used the same resolution).

I want to get that case but I heard that the new case has a bigger hole for camera and a new cutout to accommodate the change in light sensor location.

Yes that is correct.
 
I bought the old Iphone 4 otterbox universal for my 4S. Alignment is great, no water marks. I have a screen protector on aswell.
