Encrypting drives before using them as backups?

I'm going to setup some 6TB drives to use with Carbon Copy Cloner to backup various drives.

1. How do I encrypt the drives prior to doing this? Do I need to reformat them and encrypt doing reformatting?
2. Is there a way to encrypt a drive that already has 5TB of data on it, or will it take too long?
