For me, firefox 3.5.3 was somewhat slow—noticeably slower than Safari.

3.5.4 Seems to have sped up the browser a lot for me.

However, I also deactivated a few extension at the same time i upgraded.

So I am wondering...

Have any of you seen any speed improvements with Firefox 3.5.4 update?

Almost definitely the plugins. A few plugins make the difference between a snappy and a sluggish Firefox for me. My brother's Firefox was unusably slow until I uninstalled the three different search engine toolbars that some software had installed without asking him.

when i click on "Check for Updates" it tells me no updates available.

how can i fix this?

I have 3.5.3

Just download and install 3.5.4 from mozilla.com.

As for myself, I found Firefox 3.5.x to be unpleasant in terms of performance so I moved to Safari 4.0.x and find performance to be much better, especially in OS X 10.6. That being said, I acknowledge that one's experiences shall vary.
