The basic process is to use redsn0w to get into the pwned DFU mode and then use a custom activated and jailbroken firmware. You then just use ultrasn0w to unlock.

The method I used first was to start TU, save my blobs and then upgrade to 4.1 in Itunes and then use Greenpoison to JB and activate.

This new method I am unsure whether you still need to run TU.
 
You don't need TU with this method, Redsnow > DFU > custom firmware. I upgraded last night on my IP4 and 3GS while still keeping the baseband.

TU should make no difference. You are either using it to save your SHSH's or to ugrade to 4.1 and keeping your baseband. If you create a custom firmware and apply it using Redsn0w it could care less what's on your phone. It will be replaced with the custom firmware.
 
This was straightforward to say the least.

Put the phone into DFU mode using redsnow as in the link above.

Then download the custom firmware and restore.

No Need for TU.
