Getting a external HD for all my iTunes music and videos and I have some questions...

Okay I'm planning on ordering this drive,http://www.newegg.com/Product/Product.aspx?Item=N82E16822204069, to put all my media files on so I can clear up space on my MBP. Is it possible to make iTunes just read from this instead of my HDD on my Mac? Also I can partition a portion of the drive for Time Machine correct?

Yes, move your Library to the external and point iTunes to the new Library under Prefs > Advanced > Library Location.

and you can partition and have time machine use one partition.
