I don't have mobileme and I want to upgrade now.

Yeah you have to get snow leopard first before an upgrade to lion. After you install SL, go the Mac app store and buy Lion

You will need to purchase a copy of snow leopard from apple.com and THEN you will be able to upgrade to lion via the App Store. Apple Stores do not carry a copy of snow leopard OR Lion.

Can I just get a USB copy and do a fresh install (been meaning to this for ages anyways)

Yes, and the iPhone Apple Store app shows that Lincoln Park has the USB stick in stock for $69.00

Probably want to re-verify that before you head out.
