Hi,

I am wondering if the following is possible:

I am planning on installing a door sensor / vibration sensor on our front door.

Is it possible to set an automation where the sensor is enabled when the last person leaves the house and disabled again when one returns home?

how would the house know when the last person leaves? What are you using for your smart home? With Alexa you can set routines. I don't have a smart lock but if I did I could have a routine where I say "Alexa I am leaving" and it would turn off all my lights, and lock my front door. I would think something like that is the simplest.
 
I assume it knows who is logged into your wifi and when someone isn’t logged in I.e. isn’t home.

Apple offers these kind of automations but I’d like to know if door sensors can be programmed in this way through HomeKit.

Don't think I would ever want to trust sensors to know when I am there or not. When I use my voice for actions, it at least acknowledges or says something went wrong and I know it did not complete
 
I say what ever you go with Home automation don't go with Cloud based systems you hardly use! To save money go with a product uses their own storage you add (NAS) or center control hub and doesn't have a subscription to buy only a certain camera or sensor!
