I know about global spotlight but arstechnica references to the ability to search by mail specific criteria and implys it's within mail but I don't know to activate that feature.
 
First you have to have OS 3.0 Installed. After that, you open your mail account and if the search bar is not on the top then just scroll down and it will pop out.

Another thing, not all email service's support search. What are you using?
 
I didn't have to activate anything to get it.

I just went into my Inbox, and then dragged my finger down on the screen at it was up there.

Go to your inbox, scroll up you'll see the search bar. Then you can search by from, to, subject and all.
