As is, it's two click process. Go to dock > click on folder > click on open in finder. 9 times out of 10 I'm opening in finder so I figure cut to the chase.
 
Or if it's a multi-button mouse, right click on it on the dock and select "Open".

ya that's still 2 click. And I did see th keyboard short cuts but I'm more of a point and click guy.

I did just find out that if you enable tabs in finder those tabs will always stay open until you close them. Handy

The only way I have found that works on one click is to make an alias of the folder and drag that into the doc
 
I have a couple of folders I access often, all I did was create an alias of the folder and drag it to the right side of the dock. All it does is open the folder in finder though.
