What is the best way - I see the 27" iMac has 4 slots, so can I order x2 4gb modules from Crucial and install, giving me 16gb ?

Will there be any difference between four 4gb sticks and two 8gb's (speed maybe) ?
 
I'm using 2 Crucial memory sticks with the pre-installed RAM and now have 16GB running at 1600Mhz, so you'll be fine using the existing memory.
 
Yep go with two 8Gb sticks I usually just go with what apple would have done if I got that much from them. Plus it allows u too get more later without having to replace it all
