Initial Time Machine Backup Help: Local --> Airport

Hi all!

I have over 250gb worth of data to backup to my new 1.5TB Seagate external HD.

I have attempted to backup locally (via USB2.0) and then plug the HD into my AEBS for wireless Time Machine, but upon the next backup it starts all over again....

Is there a trick to backup locally on the initial run and then for subsequent backups have it over Airport?
