Looks like the new iPad will be released at 5:00PM this Friday, if you go to apple's website and look up your local store, you'll see that apple doesn't have any classes planned after 4:30PM, prior to that, the whole day is full of scheduled activities.
 
My local store has a workshop planned for 5pm - 6pm on release day.

I'm in New York this week and was hoping that the 5th Avenue store would start selling at midnight, as they are open 24/7.

I'm from NYC, but i think i would rather wait around apple's upper west side store, 5th avenue is pretty hectic, and a midnight release is unlikely, all apple stores in NYC generally have the same release time.

I'm currently working about 30 seconds from the Apple store on the corner of 9th & West 14th Street-if I stick my head out of the office window I can see it from here!

Having said that, I'll almost certainly have finished here by Friday and I'll be staying in Midtown Manhatten, so I guess my closest stores are the one you mentioned, 5th Ave and Grand Central.

If you're going to be in midtown your closest stores are 5th avenue and the one on 68th and broadway on the Upper Westside.
 
They will go on sale as early as the store opens as the Apple web site would clearly spell out 5pm (as they could be sued for inciting a riot).

This is good business sense (not causing a riot)
