I charged my iphone 4s to 100% last night, and I went to sleep.
i woke up this morning, the battery dropped 8%, the standby time is 12hours.
and the usage time is 30 mins, HOW COME? I did not use the phone.
I had turn off the notification, gps location, iclound,wifi,siri..
What else I have to turn off to solve this problem?

also 3g and celluar were all off
 
doesn't matter even if you disable all setting off and brightness 0 % battery still drain about 3 or 4 % since you standby ... that how iphone work ....
 
Clearly the OP is asking where his 30 minutes of USAGE is coming from, not why the battery was drained. No one reads anymore.

12hours eqauls about 6-7% on the battery, remember a 4S only has 200hour standby, if you count it in percent, that equals to 5% for 10hours.

I'm sure your conclusion is right, but the word "Clearly" does not describe OP's question. If the battery drain is not an issue, he shouldn't describe it in such detail.

OP, you turned a bunch of stuff off - what WAS running? Alarm clock?

Was airplane mode on? If not, try it tonight with airplane mode & see if it happens again.

It doesn't matter what it is, it won't do any difference, or maybe 1%, is that really something you wanna find out to save 1 percent of battery life? Wouldn't think so.

The battery drain he has had is to be expected and is very normal. There is no issues, so there is really no answer to his question.

OP how often is your setting to check email? And how many accounts do you have. That is almost certainly where this usage time comes from.
