#1) This ONLY shows on a black screen and it is dark blue in color.

#2) The photo was taken with my wife's iPhone 5S of a total BLACK screen on my 6 Plus and it makes the "blemish" look 100X worse than it actually is. It is visible but not terrible in "real life" but it is still there. It actually looks like a dark blue fingerprint and once again the photo makes it look MUCH MUCH worse then it actually is.

Are these dead pixels or just a screen defect? It's weird how in the photo the screen looks nothing at all like the total black it actually is but it certainly makes the spot stand out.

Take it to the Apple store, genius can just replace the front panel and not the whole device. But make a back up just in case.
