Is there a way to backup my iPhone by creating an image file of the content? If so, how?
Again, I am talking only about creating an image file, not any other backup method.

Nope, the iPhone can't be accessed like a mass storage device, so this kind of backup/restore is not doable.

I am trying to understand - there is no way that a third-party program will do the job?
If not, is there any way to backup the iPhone including the jailbreak, cydia tweaks and apps settings?
 
Jailbreaking cannot be backed up. Your applications and media files are backed up in iTunes (apps/settings into the iPhone backup file, media is stored in your library). If you want your account information (email account, iCloud account) to be part of the backup you need to set iTunes to use a password to encrypt your backup file.
