I am using Carbon Copy Cloner fyi. Can i use my mac or do i have to use my pc untill my backing up is done?
 
Browser: Meh. iTunes? Give that one a little bit of thought (but don't lose sleep over it).

Files that are open as writable while the backup is running aren't going going to get backed up in a useful state, if at all. For iTunes that means your library (the database part) may not be backed up properly (but the music and videos themselves should be okay).

For a browser, downloads in progress and cache junk would be the victims. You probably don't care about 'em.
 
I've used CCC before but with 10.4.X it went bonkers and didn't work. Now all I do is pop in my Tiger install DVD, restart and boot off the DVD. Then run Disk Utility and use the Restore tab to backup my system. This way you don't have to worry about various programs running while backing up.
